Objective: To determine frequencies distribution of peroxisome proliferator-activated receptor γ2 (PPAR-γ2) gene Pro12Ala polymorphism genotype and peroxisome proliferator-activated receptor-γ coactivator-1 Gly482Ser allele in Han and Korean population of Yanbian area in China, and relationship with type 2 diabetes mellitus. Methods: 257 people from Yanbian were randomly selected, including 70 Han nationality people and 75 Korean nationality people with normal glucose tolerant(NGT),55 Han nationality patients and 57 Korean nationality patients with type 2 diabetes mellitus (DM). PPAR- γ 2 and PGC-1 gene variant was detected using Polymerase chain reaction-restriction fragment length polymorphism .Case-contrast analysis was used to study the association Pro12Ala polymorphism between NGT and T2DM. Results: (1) The G allele frequencies of NGT subjects were 4.00% for Korean,3.57% for Han, but in T2DM patients were 2.63% for Korean, 3.57% for Han in PPAR- γ 2. and no significant difference was found between this two groups (x~2=0.127, P=0.75); There was no association with frequencies compare CC gene and CG/GG gene, also in the G allele frequencies in T2DM and NGT of two nationality. (x~2=0.188, P=0.75;x~2=0.149, P>0.05) . However, there were significant differences when compared with European White and Japanese of USA. It was lower 10%~14% than European White and 9% Japanese of USA. (2) The G allele of frequencies of NGT subjects were 49.00% for Korean, 46.00% for Han, but in T2DM patients were 51.00% for Korean, 48.00% for Han in PGC-1. and no significant difference was found between this two groups (P>0.75 ); There was no association with frequencies compare GG gene and GA/AA gene, also in the G allele frequencies in T2DM and NGT of two nationality .
