Objective: To study the expression of IL-18BP in the specimens of primary hepatic carcinoma, nodular cirrhosis, chronic hepatitis B and healthy hepatocellular tissues as well as the levels of IL-18BP in serums. The relationship was investigated between the expression of IL-18BP and the clinic pathological features of chronic hepatitis and the primary hepatic carcinoma .Methods: New Zealand rabbits were immunized by Pro-IL-18BP-N polypeptide emulsified in Freund's adjuvant to prepare IL-18BP. The expression of pro-IL-18BP were detected in the specimens of 41 primary hepatic carcinoma, 11 nodular cirrhosis, 9 chronic hepatitis B and 5 healthy hepatocellular tissues (5 cases were taken from the patients of hepatic cyst ) by the indirect immunohistochemical staining. The serum levels of IL-18BP among 25 primary hepatic carcinoma patients, 20 nodular cirrhosis patients, 20 chronic hepatitis B patients and 15 control subjects were measured by ELISA.Result: The titer of pro-IL-18BP was 1:30000~1:50000. The positive rate of IL-18BP in surrounding-tumorous tissue, tumorous tissue, nodular cirrhosis and chronic hepatitis B were 97.56%, 34.15%, 36.36% and 22.22% respectively. There was no positive expression in the control groups. IL-18BP was overexpressed in surrounding-tumorous tissue of primary hepatic carcinoma and the expression of IL-18BP in surrounding-tumorous tissue was significantly higher than the other three groups(P<0.005). The multiple comparison of the other three groups has no significant difference (P>0.05), but the nodular cirrhosis group was higher than the chronic hepatitis group in the number of positive cells and the staining intensity. The serum levels of IL-18BP in control subjects, chronic hepatitis B, nodular cirrhosis and primary hepatic carcinoma were 3.25±0.53 ng/ml, 6.20±1.48 ng/ml, 14.90±1.35ng/ml and...
