Clinical Application And Preparation Of Monoclonal Antibody Of Human Sperm Protein 22

Sperm protein 22 (sperm protein 22, SP22) is encoded by housekeeping gene of euchromosome. In rat, it is localized mainly in heart, lung, brain, live, kidney, seminiferous tubules, epididymis, testis. And SP22 expressed mainly in testis. SP22 is also expressed in various tissue of human, mainly in heart, musle, pancreas, pituitary, adrenal gland, thyroid gland and saliva gland. SP22 mRNA is transcripted in pachytene spermatocytes, secondary spermatocytes and round spermatids during spermgenesis, but the protein of SP22 only exists in late spermgenesis. As a novel protein related to fertility, SP22 not only has the activity of enzyme but also has the function of regulation factor and cancer-causing factor. The SP22 on the surface of sperm can combine with the zona pellucida of the oocyte and participate in acrosome reaction as well. Therefore there is a need to acquire large amounts of purified SP22 and its antibodies, not only for establishment of an immunological method to detect SP22, but also for exploring its function in male reproductive system and relation with male infertility.In the first part, on the basis of previous recombinat E. coli.BL21. the expression of SP22 was induced by IPTG. Then the recombinant SP22 was purified with Ni-NTA resin and identified by SDS-PAGE. Finally recombinat SP22 of high purification was obtained.
