| [Objectives]Establishing the kainic acid induced rat temporal epilepsy model, exploring the making method and investigated value of it, the objective of this experiment was to discuss the roles of the neuro-immune-endocrine network in epilepsy, by studying the change rule and function of S100βprotein and cytokines including interleukin (IL)-8 and IL-10 in epilepsy rats.[Methods]kainic acid was injected into hippocampus'CA3 of rats by stereotaxic apparatus ,and make the rat having epileptic seizure.We observed the process of epileptic seizure and the behavior change, recording electroencephalogram at the same time . We detected content of interleukin-8,interleukin-10 in serum and expressing of S100βprotein in hippocampus by radiommunoassay,enzyme linked immunosorbent assay and immunohistochemistry .[Results]Twenty eight of 50 stimulated rats were produced 5 stage seizure,and fourteen of 42 stimulated rats were produced 4 stage seizure.The right and left frontal lobe had persistence firing of spike wave or sharp wave in EEG. The experiment also found that the kindled rats were more irritable and their defense grade was higher than the control and sham groups.The level of IL-8,IL-10 in serum were begin to increase in 6h after the time when kainic acid was injected into hippocampus'CA3 of rats, reaching the peak in 12h, decreaseing in 24h, back to the normal level in 72h. The ratio of IL-10/IL-8 were begin to decrease in 6h after the time when kainic acid was injected into hippocampus'CA3 of rats, reaching the lowest level in 12h, increaseing in 24h, back to the normal level in 72h.
