| Objective:To study the anti-tumor effect and explore its mechanisms of arsenic trioxide(As2O3) combined with fluorouracil(5-FU) on the human colorectal carcinoma cell line LoVo.Methods:In vitro,human colorectal cancer cell line LoVo was treated by As2O3 and(or)5-FU in various concentrations respectively alone or in combination,and then the effect of cell growth inhibition was assayed by MTT method;cell cycles were analyzed by flow cytometry(FCM);the expression of Survivin,bcl-2,bax and VEGF(vascular endothelial growth factor)were analyzed by real-time poly merase chain reaction (Real-time PCR)and immunocytochemistry(ICC).Results:As2O3 or 5-FU could inhibit the growth of the human colorectal carcinoma cell line LoVo,and the inhibitory effect was dose-and-time dependent(P<0.05 or P<0.01).Compared with As2O3 or 5-FU individual drug group,As2O3 combined with 5-FU significantly inhibited the growth of LoVo cells(P<0.05 or P<0.01).The cell cycle distribution was altered , the ratio of G0/G1 phase cell decreased, the portion of S phase cell increased;... |
