partⅠExperimental study on the change of collagenⅠ, TIMP-1andα-SMA for liver cirrhosis induced by CCl4 in rats Objective:To observe the change of collagenⅠ, TIMP-1andα-SMA when cirrhosis induced by CCl4 in ratsMethods: Forty female SD rats were randomly divided into two groups, each group had twenty rats. They were established control group and liver cirrhosis animal model. Detected ALT. Measured body weight, ratio of between liver or spleen and body. Observed liver whole pathological status, HE stain and immunohistochemistry in rat livers. Observed hepatocytes and its interstitial substance in rats with electron microscope.Results:1. The concentration of ALT in liver cirrhosis group was significantly higher than in control group(p<0.01). 2. The body weight in liver cirrhosis group was significantly lower than in control group(p<0.01),and ratio of between liver or spleen and body in liver cirrhosis group was significantly higher than in control group(p<0.01). Five rats died in liver cirrhosis group during making models, none in control group. 3. The whole pathological status and HE stain of livers in liver cirrhosis group displayed obvious liver cirrhosis. 4. Immunohistochemistry : CollagenⅠ, TIMP-1 andα-SMA in liver cirrhosis group were significantly higher than in control group(p<0.01). 5. Observed with electron microscope: Collagen can be seen in intercellular substance, further more, in cytoplasm. Otherwise, pathological process can be seen. But none can be seen in control group.
