Study Of Integrins And Several Other Relative Inmmune Factors In The Pathogenesis Of Diabetic Nephropathy And The Intervention Effect Of MMP In Rats

Objective:To investigate the expression of integrins, OPN, TGF, CTGF in the kidney of diabetic nephropathy rats;To test the hypothesis that MMF prevent the development of DN by anti-inflammatory effects and study its pathology by immune.Methods:10 SD rats were randomatically selected from 36 to be normal controls(group A).Others were made diabetic by single intraperitonel injection of STZ.The rats whose UAER>20ug/min were selected and divided randomly to diabetic nephropathy controls(group B) and MMF treatment group(group C).Rats of group C were givin MMF 10mg/kg/d while group B given the same amount of NS.Observe the change of blood glucose,UAER.Sacrificed the rats after 2mo,and observed the change of histology and expression of integrinβ1,α5,β3,OPN,TGF-β1 and CTGF.Count the amount of tinct cells in glomerular and tubloinstetial.Results:1. Blood glucose,kidney weight/body weight(KW/BW) and 24 hours urinary protein increased significantly in DN model group and MMF treatment group,compared with control group (P<0.01).2.Histological changes of glomerular and tuberlar in DN can be observed.3. By immunohistochemistry Integrinβ1 were observed expressing both in glumerular and tubulointerstitium of all groups. In glumerular the level of group B(19.25±4.65) increased than in group A(4.80±2.89) significantly(P<0.01).There was a little decreasing in group C(16.83±4.53)(P<0.05) but still much more than in group A. (P<0.01) 4. Integrinα5 was mainly expressed in tubulointerstitium and little in glomerular.Increased level of Integrinα5 in tubulointerstitium of group B(195.92±44.61) and C (162.33±21.29)were abserved compared with of group A(14.06±6.90)(P<0.01) (P<0.01).In group C there was a little decreasing compared with in group B(P<0.05).5.
