The Osteonecrosis of the femoral head (ONFH) among which the most cases were induced by Steroid, is a common disease. It often occurs and causes disabilities in clinic. Previous studies showed that Steroid may induce marrow stromal cells (MSCs) to differentiate into adipocytes followed by systemic changes in lipid metabolism, and then osteonecrosis eventually develops. Plenty of experimentations on animals also give us the same result. But there were very few reports on human Marrow stromal cells so far.We isolated, obtained and nurtured the original human Marrow stromal cells,into which dexamethasone was added. The effects of dexamethasone on human Marrow stromal cells were observed by the methods of molecular biology to further investigate the pathological mechanism of Steroid-ONFHMaterials and MethodsThe bone marrow was offered willingly by ten inpatients of the first affiliated hostpital of Zhengzhou university, the bone marrow was cultivated and isolated by adherence after taking it in operation from iliac bone.After several passages, the primary hBMSCS were divided into two groups at random,one was the controlled group,the other was the contrast group.We added 10-7M dexamethasone into the first group and nothing into the second group.The expression of PPARγ mRNA and Osteocalcin mRNA in the two groups was detected by quantitative Rt-PCR technique 5 days later.Results1.
