Objective we take myeloma cells RPMI8226 for target cells,explore the effects of STI571 on cell proliferation, adhesion and the expression of Racl mRNA.Methods The effects of STI571 on the proliferation of RPMI8226 cells were determined by MTT assays. The effects of STI571 on the cell cycle of RPMI8226 cells were detected by Flow cytometry.Crystal violet staining was used to observe the effects of STI571 on the adhesion rate of RPMI8226 cells to fibronectin. The adriamycin (Adr) resistance mediated by cell adhesion was determined by MTT assays which were divided into four groups:①FN+Adr group;②BSA+Adr group; ③STI571+FN+Adr group;④STI571+BSA+Adr group. Racl mRNA levels in RPMI8226 cells were examined by semi-quantitative RT-PCR.Results The proliferation of RPMI8226 cells was significantly inhibited by STI571, the cell mean IC50 values were 34.52 ± 2.31 μ mol/L, 28.46±1.58 μ mol/L,25.74± 2.44 μ mol/L repectively after cultured with STI571 for 24,48,72 hours.After administrated 25 μ mol/L STI571 for 24...
