The ATP-binding cassette (ABC) transporter superfamily is mainly composed of membrane proteins that translocate a wide variety of substrates across extra- and intracellular membranes, including metabolic products, lipids and sterols, and drugs. Overexpression of certain ABC transporters occurs in cancer cell lines and tumors that are multidrug resistant, which contribute to the multidrug resistant in clinic. Genetic variation in these genes cause or contribute to a wide variety of human disorders. The precise mechanism that how to drive the transport and some function are not well understood or completely unknown, and the intra-protein interactions are poor known. Large scale research of protein-protein interaction may reveal precise mechanism of transport and function.Protein-protein interactions are the basis of all organisms and characterizing the role of protein-protein interactions is indispensable to understand life arcanum. Large scale examining the protein-protein interactions with yeast two-hybrid system reveals the function of proteins, which is highly technique-demanding rather than simple baits construction. This research focuses on optimizing key procedures of yeast two-hybrid screening of the ATP-binding cassette (ABC) transporter superfamily protein. Twenty pairs of protein-protein interactions with high reliability were identified, thus provide clues for their common transportation mechanism, find new novel functions and also to provide some experiences for large scale searching protein-protein interactions using yeast two hybrid system.In this dissertation, we use the carboxyl or amino terminal of fourteen crucial proteins of which stand for six subfamilies of the ATP-binding cassette (ABC) transporter superfamily to screen human liver library by yeast two-hybrid.
