The Experimental Study Of Hypoxic-preconditioning Effect On The Transplanted Liver Sinusoidal Endothelial Cell

Objective:ON the basis of stable rat orthotopic liver transplan -tation's(OLT) models. Donor rat admins hypoxic preconditioning in preoperative.To study the construction and function changes of transplanted liver sinusoidal endothelial cell in post-operative 2,24 and 48 hours,meanwhile ,to observe the correlation with HIF-1 expression.To investigate the effct of hypoxic preconditioning to transplanted liver.Methods:1 . SD rats were randomly divided into three groups.A:model control group,B:hypoxia graft group,C:normal control group.2.Tissue of graft livers adopted after operation were observed by electron and optics microscope. Expression of HIF-1a was examine by SP immunohistochemical method in liver.Content of HA and ET-1 in serum were detected by radio-immunity.Automatic biochemistry analyzer determine the indexs of hepatic function.Results:1.Standard of stable OLT models is able to maintain steady respiration and heatbeat, revive and turn body over operation. Cold ischemia time,unhepatic period and warm ischemia time of OLT were 50±3.15 min, 20±2.15 min and <5sec.The one week survival rate of B group is obviously supper than A group.2.ALB of A groups is lower than B group in 24h after OLT operation(P<0.05). TBIL,ALT,AST,ALP is supper than B group(P<0.05).3 .Post-operation light microscope observation display cellular edema even cell death , Liver plate is thickening, sinus hepaticus stegnosis, hepatic cell steatosis,changes of B group is lower than A.Electron microscope appears SEC fragmentation, sinus hepaticus cavity appears generous vacluolus in postoperation 2h. 24h of postoperation,SEC swelling,nuclear accrete,endoplasmic reticulum dilat,mitochondria swell and vacluolus.SEC outstanding even ablate.sweelling of B group is lower than A...
