The accumulation of neutral lipid.such as cholesterol, plays a key role in the foam-cell formation and in the development of atherosclerotic lesion. In eukaryocytes, the deposited lipid is in the form of lipid droplets. The content of adipophilin is highest among some 40 lipid droplet-related proteins In the promoter of adipophilin gene, peroxisome proliferator responsive elements (PPREs) that respond to all subtypes of PPARs, can bind PPARγ1, γ2 and further modulate the expression of adipophilin gene.What's more, nuclear receptor peroxisome proliferator-activated receptors (PPARs), which are involved in regulating the lipid-metabolism-target-gene-transcription, interact with transcriptional coactivators PPAR-binding protein PBP and then receives the direct modulation of PKC phosphorylation. To have a further study for the regulation factors in lipid-accumulation mediated by adipophilin, especially for the PKC signal transduction pathway that takes on a good pharmic prospect, may provide a new therapic strategy for atherosclerosis. Our study, firstly, observed the expression of adipophilin and PKC α in aortic atherosclerotic leisions of the New Zealand rabbit; then measured the PKC activity, the expression of PKC α , PPARγ and adipophilin, respectively, in the A10 rat smooth muscle cell, THP-1 macrophage and ECV304 human umbilicus vein endotheliocyte incubated with oxLDL for 24 hours; finally, detected whether the changes of PKC activity influence the expression of PKC α , PPARγ and adipophilin, in the THP-1 macrophage treated with PKC activator PMA and inhibitor Calphostin C for 16 hours, intracelluar lipid, also measured parallelly, to further explore the role of PKC in lipid-accumulation mediated by adipophilin.
