1. To build an effective rat experiment model of Diabetes Mellitus and to explore themost suitable dosage of STZ with intraperitoneal injection.2. To find a method for isolation and purification as well as the evaluation of islet cellquality.3. To investigate the correlation between ICAM — 1/LFA— 1 and islet cell transplantation via portal vein in rats. Try to provide the theoretical evidence for the use of mAb in clinical or research practice.Methods: the study was divided into three parts:1, Building the diabetic rat model: 24 Wistar rats were divided into two groups equally at random, the one group were injected STZ at a dosage of 55mg/Kg intraperitoneally, while the other group accepted a dosage of 65mg/Kg.2, Rats' islet cells were isolated by the solution of the collagenase V injected through the common bile duct and purified by Ficoll's discontinuous density gradient centrifugation. The identification of islet cells and alive cells were studied with DTZ and with AO and PI. Glucose -stimulated insulin secretion during subsequent static incubation was measured using the ultrasensitive rat insulin ELISA kit.3 , Approximately 800 islets from Wistar or SD rats were transplanted into the liver of STZ-induced diabetic Wistar rats via the portal vein free of mmunosuppressant. The plasma glucose and ICAM— 1 and LFA— 1 levels were monitored and grafts were studied morphologically. The duration of normoglycemia and the graft survival time were compared.Results.1,The rate of fully diabetic rats injected at a dosage of 65mg/Kg was 91.67%, zero death rate, while the rate of 55mg/Kg group was only 8.33 %, (p<0.05).2,The yield of islet cells was 400-640 islets/pancreas. The mean yield was 490 ± 90islets/pancreas. Islet purity was over 80%, islet viability was more than 90%. The insulin release test indicated the islet cells were sensitive to glucose stimulation.3 , The expression of ICAM— 1/LFA— 1 after transplantation in allo-gene graft group was significantly higher than that in syn-gene graft group(p<0.05). and the serum sugar in allo-gene graft group after transplantation early decreased to normal level, but climb up to high level above normal from 9th day.
