Capillary electrophoresis (CE) is a relative new technique of separation and quantitation regrouping different modes of separation, which has raised worldwide interest during recent years and many applications in the separation. The high efficiency, powerful resolution, fast separation, low instrumental cost, and low consumption of samples and reagents are the main advantages of CE over high performance liquid chromatography (HPLC).In this paper, we have developed three new methods with capillary electrophoresis to study trace nimesulide in human serum, to investigate pharmacokinetics of gemfibrozil, ornidazole and tinidazole in vivo, real time and on-line in rabbit coupled with microdialysis, to research into the binding interaction of rimantadine hydrochloride-human serum albumin by capillary electrophoresis with laser induced fluorescence.The thesis consists of two chapters:Chapter one: The review of capillary electrophoresis in drug analysisIn this part, a review about capillary electrophoresis, the application of microdialysis coupled with capillary electrophoresis in drug analysis and pharmacokinetics study, analytical methodology for drug and protein binding studies by capillary electrophoresis were summarized in detail according to articles in recent years.Chapter two: Research reports which are composed of four components as follows:1. Study of nimesulide in human serum by capillary electrophoresis separation with UV detectionIn this study, a simple, sensitive and specific separation and analysis method has been developed for the determination of nimesulide. Nimesulide (methanesulfonamide, N-[4-nitro-2-phenoxyphenil]) was determined by capillary zone electrophoresis (CZE) in human serum. The types, pH, concentration of background buffer, the voltage of separation and injection time were optimized.The separation was carried out on a 47cm (40cm to the detector) ×50μm i.d. fused-silica capillary.
