Focus: This article use the MMP inhibitors which we foundto test their effects on the adhesion, spread, invasion andproliferation in the tumor cells. All of these experimentsprovide a strong cytological proof to confirm the results ofenzyme kinetics and a basement of further research on themechanism of the inhibitors.Method: We use the HT1080 cell which could secret severalMMPs. HT1080 cell was cultured and counted aftermicroscopically photograph to quantify the adhesion;theexpansion of cell was measured on/in the collagen I;cellmigration assay used wound healing model, and the cellinvasion assay used the transwell invasion chambers whichhave been embedded with matrigel and NIH-3T3 cell culturemedia as chemotactic factor;cell proliferation assay usedMTT assay.Result: In vitro experiment indicated that the compound Gcould effective inhibit cell adhesion at 10uM;the compoundA could depress the spread of cells cultured on the collagen I,and compound D could inhibit the spread of cells in 3Dculture;compound A, D and F all display the ability ofinhibition in the adhesion assay;statistical analysis indicatesthat both the compound A and D could influence the tumorcell invasion;the IC50 of compound A, D, F, G wasmeasured by the MTT assay.Conclusion: After the use of the four inhibitors in cell, wefound that all the inhibitors have different effects on the celladhesion, spread, invasion and proliferation. BecauseHT1080 express the MMP-1, -2, -3, -7, -9, -14, -15 and -16and the inhibitors we used can inhibit the activity of MMPs.Furthermore, for MMPs involve in many behaviors of tumorcells, our reasults provide a proof of these tumor inhibitorson the cytobiology.
