| Objective: To research the relations among interleukin-1α(IL-1α)-889C/T polymorphism,the polymorphism of variable number of tandem repeat(VNTR)of interleukin-1 receptor antagonist(IL-1ra) gene and tumor necrosis factor-α(TNF-α) promoter polymorphism with Alzheimer's disease (AD) in Han people of ShanXi province.Methods: The gene polymorphisms of apoE,IL-1α,TNF-αand IL-1ra genotype were examined in 55 AD cases and 59 controls by means of polymerase chain reaction(PCR) and restriction fragment length polymorphism (RFLP).Result: Alleleε4 of the ApoE increased the risk of Alzheimer's disease in the Han Chinese( OR=6.497,95%CI(2.151-19.619), P=0.000 ) , and Alleleε3 was a protective factor(OR=0.401,(0.200-0.801), P=0.009). No significant difference was found in the genetype and allele frequency of IL-1α,IL-1Ra and TNF-αbetween the both groups. The presence of the ApoEε4 allele did not influence their genotypes or allele distribution. All the subjects were stratificatied by age, we found that there was significant difference in the allele frequency of ApoEε2,ε3,ε4 between the AD cases and controls groups(>70 years old) (χ2=14.983,P=0.001); while no significant difference was found in two groups(≤70 years old). Alleleε4 of the ApoE increased the risk of Alzheimer's disease when patients were beyond 70 years (OR=20.4, 95%CI (2.637-157.816) P=0.000). But we didn't found the significant difference of three proinflammatory cytokines gene polymorphism in the stracification of age between AD and control groups (P> 0.05).Conclusion: Alleleε4 of the ApoE is a major risk factor of sporadic AD, and it has correlation with age, Alleleε3 is a protective factor of AD. The presence of IL-1α(-889)T allele,TNF-α(-308)G allele cannot be used for distinguishing AD from controls; IL-1Ra*2 allele is not a protective factor of AD in Han people of ShanXi province. |
