Objective:This study was designed to (a)investigate the expression of CDX2,PTEN and ki-67 in colorectal carcinoma,intestinal adenoma and normal intestinal mucosa. and (b) evaluate the correlations among CDX2,PTEN,ki-67 and DNA ploidy and the clinicopathological parameters. and(c) analyze the effect of CDX2,PTEN,ki-67 in the occurrence and development of colorectal carcinoma in order to tell tumor malignancy, bad prognosis ,as well as provide some theoretical basis for the clinical diagnosis.Method:Fifty samples of human colorectal carcinoma tissue ,twelve samples of intestinal adenoma and ten samples of normal intestinal mucosa were examined for CDX2,PTEN as well as ki-67 by immunohistochmistry,and Flow cytometry was used to detect the status of DNA ploidy in colorectal carcinoma.Result:The expression of CDX2 in colorectal cancer, intestinal adenoma and normal intestinal mucosa were 82 %( 41/50), 91.7%(11/12) and 100 %( 10/10) respectively. The expression of CDX2 in colorectal carcinoma has significant difference compared to that in normal intestinal mucosa and in intestinal adenoma.PTEN in colorectal carcinoma,intestinal adenoma and intestinal mucosa tissue were 64%(32/50),83.3%(10/12) and 90%(9/10) respectively,the expression has significant difference.CDX2 and PTEN have both related with tumors TNM stages,tissue grading,lymph-node metastasis and primary focus infiltrating depth. Coefficient of product-moment correlation is 0.406。CDX2 and PTEN expressions were negatively correlated with the expression of ki-67 (r=-0.684, p<0.000; r=-0.511, p<0.000) respectively. The expression of CDX2 was increased in diploid colorectal carcinoma; as compared with that in heteroploidy colorectal carcinoma (93.75% and 76.47%).But the expression of PTEN has no difference in the two sets.Conclusion: 1,There was a clear positive correlation between CDX2 expression and PTEN protein in part of the colorectal carcinoma; The more tumors TNM stages is late,tissue grading is low and primary focus infiltrating is deep, the more the two protein expression is low or short; Compared with that in lymph-node metastasic colorectal carcinoma, CDX2 and PTEN expression is higher in that without lymph-node metastasis.2,Both CDX2 and PTEN expression negatively correlate with ki-67; CDX2, intestinal-specific homeobox genes, regulate cell proliferation and differentiation in the continuously renewed intestinal epithelium. PTEN positively regulates CDX2 gene expression.
