This study carried out an overall study on preparation technics, biologic activities, pharmacodynamics and toxicity of a new biological preparation which is chiefly used to treat diseases of respiratory system as an expectorant. This preparation contains several nucleases extracted from Bimastus which have such activities of as DNase, RNase, protease, lysozyme and so on. According to the results of the experiments, this preparation is safe, effective and less-cost. This study has laid experimental basis for further clinical trials.1. Studies on preparation technicsTo get an efficient, stable and time-saving preparation technics path, several programs have been tried. The best path can retain the biological activities of the active ingredients to the best extent.2. Studies on physical and chemical properties, biological activities and quality standard of this expectorantTo further understand the preparation's physical and chemical properties, many stability tests have been done. The results showed that the preparation has strong resistance to extreme acids and alkalis, alcohol and other common biological active substances (such as protease K, RNase A and lysozyme).To explore the mechanisms of the actions of the preparation, the biological activities were also been measured. According to the results, the most abundant component is DNase, at the same time, several other enzymes are also contained in the preparation. They cooperate with each other to exert the Pharmacological Actions.Based on the experimental results above, the preparation's quality standard draft was made.3. studies on pharmacodynamics The results of activity study showed the expectorant is a multi-component preparation. It definitely has several effects on treating respiratory diseases including eliminating expectoration, relieving cough, inhibiting the expression of inflammation factors in lung and killing some common bacteria and viruses in respiratory tracts.4. studies on toxicologyTo access the safety of clinical application of the preparation, Acute toxicity test, long-term toxicity test and special toxicity test have been done. The experimental results showed that the preparation is safe. It provided enough experimental basis for clinic trails.
