Objective: To investigate the effect of the combination of wild type p53 (wt-p53) gene transfected with and 5-fluorouracil(5-FU) on the growth of nasopharyngeal carcinoma cell lines CNE1 and CNE2. Methods: CNE1 and CNE2 cells that were transfected with plasmid pUHD10-3-p53 recombinant by Lipofectamine-mediate were cultured in medium contained 5-FU. 3-(4,5-Dimethy-2-thiazolyl)-2,5-diphenyl-2H- tetrazolium-bromid (MTT) technique was used to analyse the growth of CNE1 and CNE2 cells. The apoptosis of CNE2 cells was evaluated by flow cytometry. The expression of p53 gene was detected by RT-PCR method. Results: After transfected with wt-p53 gene,47.14% of CNE1 cells and 45.90% of CNE2 cells were inhibited at fourth day. And 84.74% of CNE1 cells and 85.82% of CNE2 cells were inhibited, when the cells were transfected with wt-p53 gene and exposed to 5-FU for 4 day. The apoptosis rates were 14.6% in control group, 20.7% in pUHD10-3-p53 without Lipofectamine- mediate, 25.9% in pUHD10-3 transfection group, 35.2% in pUHD10-3-p53- transfection group, 33.8% in 5-FU group, and 53.9% in pUHD10-3-p53- transfection plus 5-FU-treatment group. The high level of p53 expression was observed in pUHD10-3-p53 transfection cells. Apoptosis cells increased under fluorescence microscope. Conclusions: The growths of two nasopharyngeal carcinoma lines were obviously inhibited by the combination of wt-p53 gene and 5-FU. Apoptosis of CNE2 cells could be induced by both wt-p53 gene and 5-FU, and the effect was more obvious when CNE2 cells were treated by the combination of wt-p53 gene and 5-FU.
