Objective:To explore the characteristic of telomerase activity of peripheral blood T lymphocytes in the patients with myasthenia gravis.Methods : The subjects were 16 MG patients and 16 healthy controls . After medical history investigation and physical check-up,the test group was drawn blood of twenty milliliters by heparin resisting concrete. Peripheral blood mononucleate cells(PBMC) were isolated on lymphocytes separation medium by desity-gradient centrifugation.CD4~+T lymphocytes and CD8~+T lymphocytes were purified from PBMC by means of magnetic beadcoupled antibodies and determined telomerase activity of lymphocytes with telomeric repeat amplicfication protocol (TRAP) assays by polymerase chain reaction-based enzyme immunoassay (PCR-ELISA).Results :(1)measure of cell viability :①The percentage of CD4~+T lymphocytes and CD8~+T lymphocytes viability before and after isolation in MG patient was (98.72±1.97)% and (97.56±2.78)%,(98.56±2.31)%(P>0.05),respectively,they were no difference in cell viability before and after isolation ;②The percentage of CD4~+T lymphocytes and CD8~+T lymphocytes viability before and after isolation in healthy controls was (97.31±2.80)% and (96.94±3.73)%,(97.63±3.77)%(P>0.05),respectively.they were no difference in cell viability before and after isolation.
