Objective To make sure whether ricombinant insulin-like growthfactor can stimulate proliferation after transfecting human retinalpigment epithelial cells.Methods Human retinal pigment epithelial cells were lipofected invitro with plasmid vector pcDNA3.1:IGF-1, which encodes anepitope-tagged GFP fusion protein and a selectable neomycinresistance gene. Transduced cells were cloned in G418 and expandedfor analysis of IGF-1 transgene expression and its effect on the cellproliferation. The expression of the IGF-1 transgene in cloned cellswas confirmed by fluoresence microsrope. The growth characteristicsof transduced clones were compared with the control by trypan blueexclusion and flow cytometric cell proliferation assays.Result Cloned retinal pigment epithelial cells expressed the IGF-1transgene and transduced clones enhanced ability to proliferatecompared with the control.
