Along with the progress of the society, the exaltationof living standard, incidence of diabetes go up year byyear.It have become one of the four greatest diseaseseswhich affect the health of the mankind's in 21 centuries.Currently there has been the diabetic patients of 190million or so in the world.It will reach 330 million in 2025.There are 60 million diabetics in our country and type 2diabeteses have 95% above. The diabetes complicationsmainly cause the people's death and physically disableds.Diabetic vasculopathy includes a lot of aspects. Matrixmetalloproteinase(MMPs) play a part in the pathologicphysiology processes of extracellular matrix(ECM),such asdynamic equilibrium, tissue remodeling, reparation etc.Make use of intervention study lowering the outcomeactivity of MMPs,attributting to suppress the developmentof diabetic vasculopathy.MMP-2(gelatinase A ,Mw 72kD)was measured in type 2diabetic patients with vascular disease and normal controlsubjects,to explore the association between MMP-2 anddiabetic vascular disease.The members of case group came from type 2 diabeticpatients in the Second Clinical Medical College of JilinUniversity from September 2003 to September 2004, all werethe endocrine and cardiovascular section inpatients,male75,female 45, the average age 62 years old. The calculatorscaned the white zone, using the NIH Image1.57 softwarecalculation density.The experiment datas were adopted theMicrosoft Excel5.0 software to carry on the processing,group comparison was adopted Student-Newmen-Keuls test.We found the activated MMP-2 in diabetic patients withcoronary heart disease increases to 223% than that of innormal control subjects.However, the activited MMP-2 ofthe diabetic patients with nephropathy decreased by 30%.MMPs are a family of structurally and functionallyrelated zinc-dependent endopeptidases consisting of atleast 30 enzymes which are able to degrade all the proteincomponents of the extracellular matrix.The ECM is used as the main composition of the bloodvessel wall, playing an important role in the diabetes withcoronary heart disease and diabetic nephropathy.In thisexperiment, the diabetes merge to compare with normalperson, the expression of the serum MMP-2 in the diabeteswith coronary heart disease is high 223% than the healthy.This result further proved an important function of theMMP-2 in the atherogenesis.MMP-2 can particularlydegradation the collagen protein and basement membrane,accelerating the disruption of plaque and this also is amain reason of diabetic patients merging the myocardialinfarction. The results showed that the activited MMP-2 ofthe diabetic patients with nephropathy decreased by 30%.We considered the expression of MMP-2 decreased inExtracella Matrix(ECM) of diabetic nephropathy.That we adopt SDS-PAGE zymography to detect serum MMP-2in the diabetes with coronary heart disease and diabeticnephropathy is the first still in the domestic, because thezymogram experiment can distinguish the contents ofzymogen and zymase.Unique characteristic of the MMP-2 isonly activity form can produce a marked effect indegradation of ECM.So examination the activity of zymaseseems to be the meaning graveness in the diabetes withcoronary heart disease and diabetic nephropathy.The MMP-2 expression abnormality is one of thedangerous factors of the diabetic vasculopathy. Theresults showed that significant increasing of serum MMP-2in the diabetic patients with coronary heart disease ,whileMMP2 tended to decline in patients with diabeticnephropathy. So the relationship of MMP-2 and diabetes isvery close.Therefore examine the change of the serum MMP-2not only contribute to discover of earlier period ofdiabetic early treatment, but also have the bigger meaningto early treatment of early treatment.
