Object Effects of microinjecting substance P and its selective NK-1 receptor Antagonist(N-acetyl-L-tryptophan3,5-bis(trifluoromethy)-benzylester,NALT BE) into lateral cerebral ventricle on sleep-wakefulness cycle and emotional behavior in rats were investigated. Immunohistochemical method was used to realized c-fos in the brain to revealing the mechanism of SP effect on wake-sleep cycle.Methods 80 adult male Sprague-Dawley rats were divided into 2 experimental processes: polysomnography (PSG) process and emotional behavior process .Rats of polysomnography (PSG) process were anesthetized with chloral hydrated (350 mg/kg, i.p.), prepared for aseptic surgery, and secured in a SN-2 stereotaxic frame. Three copper cortical electroencephalogram (EEG) electrodes were screwed into the skull (one on the left and two on the right, fronto-occipital derivation). Two silver electromyogram (EMG) electrodes were placed under the nuchal muscles. One 23-gauge stainless steel guide cannulaes were then lowered to 1.0 mm above the LV (lateral cerebral ventricle) (anteroposterior, -0.8 mm; dorsoventral,-4 mm; and mediolateral, + 1.5 mm with reference to the bregma)...
