The Zedoary Turmeric Oil (ZTO) is extracted from traditional Chinese medicine, Zedoary Rhizome. The pharmacology study showed that the ZTO has many effects such as anti-tumor, anti-inflammatory, antibiosis etc. However, ZTO is a water-insoluble oily drug, its bioavailability is quite low, the disagreeable taste leads to not easy to take orally. Besides, its stability isn't satisfied and anaphylactic response and hemolysis often happen in clinical application. For these reasons, the application of ZTO was limited in some degree. At present, the liposome was taken as a good carrier of anti-tumor drug because of its many benefits. Firstly, it can increase the stability of drugs and decrease the side effects of drugs. Secondly, it can inhibit immune reaction and allergy. In addition, the liposome could cause the drug to release delayedly or targetly. The objective of this article is to improve the stability and the curative effect, to decrease the anaphylactic response and increase the safety of ZTO.The ZTO liposome was prepared by thin-film vesicle method (TFV). The method of determination was adopted by visible-spectrophotometry. The unentrapped and entrapped drugs were separated by sephadex gel and high-speed centrifugation respectively, and then the entrapped efficiency was calculated. The sterilization of ZTO liposome was used in flowing steam to last 30 minutes at 100℃ To study the optimal technical condition, we used orthogonal design. The results indicated that the entrapped efficiency of liposome could achieve 85.3%, which was prepared by using the optimized technical condition (PC/CH mass ratio =3:1;drug/lipid mass ratio=1:1;the ultrasonic time is 30 minutes;the concentration of phosphate buffer saline is 0.5mol/L). After sterilization, the entrapped efficiency of liposome was 80.2%.The physico-chemical property and basic stability of ZTO liposome were studied. The ZTO liposome was small unilamellar vesicles (SUV), round or ellipse. Theparticle diameter distribution is from 190nm to 450nm. The mean geometric diameter is 289nm. After accelerating, illumination and reservation tests, the results showed that the ZTO liposome could increase the stability of ZTO. All indexes were stable in four mouths. However, due to the instability of materials, the ZTO liposome was suitable to be preserved in conditions of low temperature and protected from light and oxygen.The hemolytic,blood vessel stimulus and toxicity tests of the ZTO liposome were conducted. The results showed that the ZTO liposome has no hemolysis and stimulus to blood vessel at administer dosage and no toxic reaction at the maximum administer dosage (240mg.kg1) on mice. These studies indicated that the safety of ZTO liposome was increased after entrapped by liposome. The anti-tumor test showed that at high-dosage (ISOmg.kg1), middle-dosage (lOOmg.kg"1) and low-dosage (50mg.kg"'), the ZTO liposome has significant inhibit effects on mice that was infected from cell of liver cancer H22(p<0.01). Therefore, the test illustrated the anti-tumor effect of ZTO liposome was increased after entrapped by liposome.
