Objective: To observe mobile changes of Nestin and basic fibroblast growth factor (bFGF) expression in hippocampal after cerebral ischemia and investigate possible mechanism of intrinsic neurogenesis.Method A total of 40 Sprague-Dewley(SD) rats were divided into control group(4), experimental groups(36) randomly.Experimental groups were divided into 3, 6, 12, 24hours and 3, 7, 14, 21 , 28 days after reperfusion. The model of experimental ischemia was made by 4-VO. The specimens were made into paraffin section. The expression of Nestin and bFGF in hippocampal zone were detected by immunohistochemistry method .Results (1) Nestin detectation: No Nestin positive cells were observed in hippocampal zone in control group. The expression of Nestin started at 3h in SVZ not in hippocampal zone and it started to increase at 6h after cerebral ischemia and reach a peak at 14d after cerebral ischemia in hippocampal zone, then decreased at 21d,28d. The difference of Nestin expression among the adjacent givengroups is significant, (p<0.05). (2)bFGF detectation: A few bFGF positive cells were observed in hippocampal zone in control group. The expression of Nestin started to increase at 3h in hippocampal zone after cerebral ischemia and reach a peak at 7d after cerebral ischemia in hippocampal zone, then decreased at 14d,21d. At 28d it decreased to normal level.The difference of bFGF expression among the adjacent givengroups is significant, (p<0.05).Conclusion(1) The expression of bFGF and Nestion in hippocampal zone after cerebral ischemia increased, which may be a beneficial response of cells to the ischemic injury. Protective funct ion(2) The sequence of expression of bFGF and Nestion in hippocampal zone after cerebral ischemia is relevant. b-FGF may promote expression of Nestin.
