Separation and analysis of active components in traditional Chinese medicine is playing the vital role in the modernization of traditional Chinese herb medicine. Because of shortage of standards, analytical methods and quality control indexes, the products of traditional Chinese medicine have had difficulties to enter international market in a long time. The modern chromatographic separation and analysis technologies have been used to solve the problem that existed in quality control of traditional Chinese medicine. In this article, macro-porous adsorption resin chromatography technology, preparative high performance liquid chromatography technology, high performance liquid chromatography technology and high performance liquid chromatography-mass spectrometry technology have been used to separate and analyze active ingredients in several traditional Chinese medicines.The studies in this thesis are summarized as follows:1. In order to purify and separate magnoflorine and laurifoline in Zanthoxylum americanum, preparative HPLC and macro-porous adsorption resin chromatography technologies have been used to purify and separate magnoflorine and laurifoline. Conditions of separation preparative HPLC and macro-porous adsorption resin chromatography have been optimized. Macro-porous adsorption resin chromatography technology firstly used to remove the salt that existed in the product. Magnoflorine and laurifoline can been used as standards to control quality of the product.2. A HPLC method for simultaneous determination of resveratrol, emodin, chrysophanol, physcion in root of Polygonum cuspidatum and its extract has been established. The method is accurate, simple and reliable. It can be used for quality control of Polygonum cuspidatum and its extract.3. A novel method of simultaneous analysis and identification of eleven flavones in the medicinal plant of Scutellaria baicalensis using high-performance liquid chromatography (HPLC)-diode array detection (DAD)-electrospray mass spectrometry has been developed. Eight components could be quantified by using external standard method with UV detecting at 276nm. The samples analyzed contained roots, leaves, stems and extracts of Scutellaria baicalensis. The method can be used for quality control of extracts of Scutellaria baicalensis.4. A HPLC-ESI-MS method for simultaneous determination of flavones in seeds of the medicinal plant Oroxylum indicum has been established. Two components could be quantified by using external standard method with UV detecting at 276nm. The method is accurate, reliable. It can be used for quality control of Oroxylum indicum.
