Objective To study the possibility and cardioprotective effect of targeted ultrasound delivering VEGF165cDNA gene and monoclonal antibody to interceller adhesion molecule-1 to myocardial ischemia-reperfusion injury of rats. Methods The research will make the gene of VEGF165cDNA and monoclonal antibody to interceller adhesion molecule-1, establish the the rat models of myocardial ischemiareperfusion injury. The experiment is divided into three part. The instrument is color doppler ultrasonic diagnosis instrument of Acuson Sequoia 512 with 4V1 probe, using twice harmonic pattern leave frequency and accept frequency 1.75 MHz and 3.5 MHz each other, adjust mechanical index to the biggest, focusing depth to be 4 cm. Trigger once per 6-8 cardiac cycle with the electrocardiogram about 10 minutes. 5% sonicated human albumin microbubble of attached with VEGF165cDNA and monoclonal antibody to interceller adhesion molecule-1 was infused into the tail vein of rats with or without simultaneous ultrasound destruction microbubbles as first and second group. The third group used as control group. Every group was divided into two groups again. One group(24 h group) were killed after 24 hours and another group(14 d group) were killed after 14 day and examined for VEGF protein expression by immunohistochemical techniqe.Microvascular density (MVD) and Polymorphonuclears (PMN) were counted in the infarcted area. The area of myocardial necrosis was measured and the experiment data means with the (?)±s, the each compare adopts square analysis.Results In 24 h group the infiltration of PMN and the area of myocardial necrosis received ultrasoundmediated is lower than others group.In 14 d group VEGF protein expression andMVD were higher in group which were received ultrasound mediated microbubble destruction for delivering VEGF,65cDNA gene and monoclonal antibody than without ultrasounmediated group and in controls. Conclusion Targeted ultrasound can increase the dencity of VEGF165CDNA gene and monoclonal antibody to interceller adhesionmolecule-1 in the area of myocardial ischemia, decrease the infiltration of PMN, reduce myocardial ischemia-reperfusion injury, improve myocardial ischemia and reduce the area of myocardial infarction.
