Liver cancer recently becomes partial curable from an untreatable disease. Its five-year survivor rate is improved to 20.6 percent from 2.6 percent. However, the cause of liver cancer so far is still elusive. As believed by most of people, it is the result of many factors, such as B hepatitis virus, C hepatitis virus, water pollution, deficiency of some trace elements, heredity, drunkenness, and so on. This article is about the research on the anti-tumor effect of Ursolic Acid developed by Li YuanHeng Medicine Company, especially about test of pre-clinical inhibitory on tumor cells and test of main medicine effects on live animal models with the tumor.1. Test of inhibitory on tumor cellsTo observe the anti-tumour effect of Ursolic Acid in vitro, we select totaling fifteen cell lines, including human colon tumor cells (HCT-8), human lung gland cancer cells (A549), human promyelocytic leukemia cells (HL-60), human cervix cells (Hela), human melanin cells (MV3), human stomach cancer cells (BGC-823), human gigantic lung cancer cells (801-D), human ovary cancer cells (A2780), human buccal epithelia cancer cells (KB), human kidney cancer cells (Ketr-3), human breast cancer cells (MCF-7), human non-small-cell lung cancer cells (PAA), human esophagus cancer cells (CaEs-17), human prostate cancer cells (PC-3M) and human liver cancer cells (Bel 7402). Employ 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) to dioxide. The research on main pharmacological effectsBy course of transplanting anti-tumor active test on tumor stems of 3LL, L1210, S180, H22, human liver cancer Bel--7402 and SMMC--7721, and evaluating the effects of Ursolic Acid on tumor cells from different sources, to provide a test foundation for clinical experiments of this medicine.Utilize wistar rats as laboratory animal subjects. They are set to five groups: normal control group, cyclophosphamide positive control group, high dose group, medium dose group and low dose group. Each group is treated in pre-settled time every day. Equal saline water was given to normal controlgroup daily during the consecutive fourteen to twenty-eight day period. Regularly measure the body weight of animals and the volume of tumors in the period of medicine given. After the experiment is over, take out the tumors to weigh, calculate the inhibit rate of tumor. Repeat the experiment 2-3 times for every tumor stem.In the period of medicine given, animals' body conditions usually do not have obvious change. Bite and sup are normal. Body weights are gained comparing to that before experiment. The volume and weight of tumors decrease after being treated with Ursolic Acid. Except L12io, its tumor inhibiting percentage ranges from 38% to 51.85%, especially to H22, Bel 7402 and SMM-7721.It suggests that Ursolic Acid has apparent anti-tumor effect upon H22, Bel7402 and SMMC--7721. |
