Objective: To investigate the effects of Carboxymethyl pachymaran (CMP) on the difference of IL-12 production by spleen monocyte-derived dendritic cells (DC) and IL-10, IFN-γ production by T lymphocyte in mixed lymphocyte reaction(MLR) in HBV transgenic mice.Methods: Spleen lymphocyte of HBV transgenic mice were incubated with different doses of CMP in vitro. MTT assay was used to detect cell toxicity of CMP. Spleen monocyte-derived DC in HBV transgenic mice were cultured with granulocyte-macrophage colony-stimulating factor (GM-CSF)and interleukin-4(IL-4). IL-12 released by DCs and IL-10 and IFN-y produced by T cells in mixed lymphocyte reaction(MLR) were measured by ELISA.Results: 1.CMP with 0-500 μg/ml were innoxious to HBV transgenic mice and normal mice . Lymphocyte proliferation index of HBV transgenic mice rose tiptop at 125 μg/ml. 2.The level of IL-12 (236.00±43.21) pg/ml produced by DCs of CMP group in HBV transgenic mice was higher than that of control group(24.67±7.51) pg/ml (P<0.01) .However, the difference was not significant between CMP group and IFN group (291.00±47.79) pg/ml (P>0.05) ; 3. The level of IL-10 (24.67±8.62) pg/ml of CMP group produced by T cells of MLR in HBV transgenic mice was lower than that of control group (74.00± 16.09) pg/ml (P<0.05). However, the difference was not significant between CMP group and IFN group (40.33±l6.26) pg/ml (P>0.05) 4. The level of IFN—γ ( 76.67± 11.72 ) pg/ml of CMP group produced by T cells of MLR in HBV transgenic mice was higher than that of control group (25.00±7.55) pg/ml (P<0.01) , lower than that of IFN group (111.33±16.56) pg/ml (P<0.05);5.The level of IL-12 was positively correlated to that of IFN—γ in HBV transgenic mice group or normal mice group (r=0.9658, P=0.000; r=0.9113, P=0.002) .There was no correlation between IL—12 and IL—10, IL—10 and IFN—γ (P>0.05) .Conclusion: CMP can significantly enhance the level of IL-12 produced by DC and IFN-y produced by T cells of MLR and inhibit the level of IL-10 produced by T cells of MLR in HBV transgenic mice. The level of IL-12 is positively correlated to that of IFN-y.
