Abstract/Summary:
A water soluble crude polysaccharide is extracted from the water-boiled dry Stellera chamaejasme, and its content is 5.6%.After graded by freezing and purified by the combined deprotionization methods of enzyme and sevage, and purified by Sepharose CL-6B gel filitration, two fractions were obtained: SP1 and SP2. They were homogeneous by tests:Gel filtration on Sepharose CL-6B,Determination of specific rotation and HPLC analyse, Cellulose acetate pellicle electrophoresis.The average molecular weight of SP1 is 226kd, the average molecular weight of SP2 is 76kd by means of HPLC. SP1 contain Rha Ala Glc Man, in molar ratio of 2.43:3.3:2.16:1 by G.C. SP2 contain Rha Ala Glc GalA, in molar ratio of 1.37:1.03:1:1.Partial hydrolysis with acid,Periodate oxidation,Smith degradation, I.R. were used on the structure analysis of SP1. The results revealed that the main chain of SP1 is mainly made up of Glc, Man; The side chains are made up of Rha, Ara, Glc, Man; The core of main chain of SP1 is made up of Glc, Man; The nonreduced end is composed of Rha, Man.Partial hydrolysis with acid,Periodate oxidation,Smith degradation, I.R. were used on the structure analysis of SP2. The results revealed that SP2 is a kind of lowly branched structure; There are majorα-configuration and minorβ-configuration; The main chain is mainly made up of Glc, GalA, there are (1→6)Glc, (1→4)Glc; The branches are made up of GalA(1→3,6); There is 1 branches among 10 main chain residues; The side chains are composed of (1→5)Ara, (1→6)Man; The nonreduced end is composed of Glc.
