Recent years, the quantity of immune injured parasitifer isincreasing, such as tumor, autoallergic disease, organ transplant,diabetes mellitus, acquired immunodeficiency, etc. In the clinic, thefungous infection shows the ascensusing tendency. At the same time,the category of eumycete pathogenic bacteria is changing persistently.The new pathogenic bacteria is increasing, though, the westernmedicine cost is expensive, and have great poison and side effect. Sothere are many clinical observations about treat this disease with thetraditional Chinese medicine and drugs, but there is no report about itsmechanism of action. Through clinic and empirical studies, It isproved that the CKJC has the a lot of preponderances, such as the lowcost price, the broad medicinal herbs resource, and easy artwork andproduce, and no manifest poison and side effect, and not easy torecidivism. The CKJC work direct to the fungous DNA, then achievethe objective of bacteriostasis. Our study provid the science fundation,and proved its scientificity and feasibility for the Chinese formulatedproducts used to cure the superficial mycosis of the skin. So it isdeserved generalizing application. The CKJC is come from my tutor—Guo-jing Yang associateprofessor's clinic work experiences for many years. It is mainlycompatibility with the drugs of anthelminthic, driving the wind evil,moistening the skin. The superficial mycosis of the skin is a kind ofcmmon ailment,frequently encountered disease in the clinic. Andthere is short of western medicine with the exact therapeutic effect andlittle side effect. For this disease, there are many using of thetraditional Chinese medicine and drugs. Beause it has the features ofthe agility making formulae, and poisonous, and its low price, it isaccepted by many people. But there is few studies about mechanismof action of the traditional Chinese medicine. Our topic depoymultitude kinds of fungous molecular biological verification methods,such as AP-PCR, a kind of new techniques which makes use of rancomposite single oligonucleotide primer to have PCR reaction toamplify the target cell DNA and the production gel electrophoresis,and analyze the polymorphism of the DNA part's size or quantity, andthen compare the genome difference. This technique can be used toidentigy the different kinds of pathogenic microorganism, includingmycetes. On the base of the above theories, our experiment used theAP-PCR to identify the mycetes strain, and observed different effectof different dosages of the CKJC, and approach its mechanism ofaction, and provid the science fundation for its therapy of thesuperficial mycosis of the skin.We random assort several category mycetes into the blank controlgroup,the negative control group,the positive control group and theCKJC large doses,middle doses and small dosage group. And then,the cultivated mycetes strain is transplanted to the surface of thenutrient medium containing the milk white moisture hand frost,theMcronasole and the CKJC in different concentration. Every 3 or 5days, we observe the effective of bactertasis. After one month, weprosecute statistical treatment and analyses, and compare thedifference between the interclasses and the effective. We respectiveabstract the mycetes DNA for both before and after bacteriostasis.Then run arbitrarily primed PCR with the random primer—OPAA11and OPD18. Then we run the electrophoresis analyses, and comparethe change of the interclass DNA band forms for both before and afterbacteriostasis. The bacteriostasis result display that the blank group ofseveral kinds of strain have no evident different with the moisturehand frost group (P>0.05);the 5% CKJC group has evident differentwith the blank group(P<0.01);the 10% and 20% CKJC group haveno evident different with the Mcronasole(P>0.05);the 10% CKJCgroup has evident different with the blank group(P<0.01);the 10%CKJC group has evident different with the 5% CKJC group(P<0.01)。Through the AP-PCR, the result shows that, using OPAA11 andOPD18 amplification, the dribs and drabs roughly homology of theblank control group,the negative control group,the positive controlgroup. The result of 5% CKJC has no difference with the dribs anddrabs before bacteriostasis. The result of 10% CKJC is entirelydifferent with with the dribs and drabs before bacteriostasis. Then weproved that the bacteriostatic mechanism of action of the CKJC is dueto change the fungus DNA. Summarizing the above, we can havesome conclusions that the CKJC have the satisfactory therapeuticaleffect for the superficial mycosis of the skin, and the CKJC of the10%,20% have the equivalency effect with the Mcronasole(P>0.05).The CKJC'S mechanism of action is to change the formation of theDNA of the mycetes, and then achieve the effect of bacteriostasis.Above all, Its mechanism action is completely diffirent with theMcronasole. The mechanism action of Mcronasole is mainly byinhibiting the biosymthesis of the mycetes ergosterin, and influencingthe permesbility of the cell membrane, so having the affection ofvacteriostatic action. Though the mechanism of actions is diffirent tothe two ones, but they have the aequales bacteriostasis action.
