Opium has long been applied for pain relief. However addictive property ofopioid alkaloid made heroin and heroine abuse a worldwide problem of medicalscience and society. Mechanisms about addiction , endurance and withdraw ofopioid have been studied comparatively deep-going, while study on opioid's effecton peripheric tissue is scare. The purpose of our study was to analyse the differenceof gene expression of adenosine deaminase (ADA) and Xanthine Oxidase(XO) indifferent regions of rat brain through researching on enzyme activity and mRNAcontent analysis of adenosine deaminase (ADA) and Xanthine Oxidase(XO)inwhich is the key enzyme in purine nucleotide catabolism . The study alsoanalysed the difference of gene expression of μ-R in different regions of rat brain.In this study,10 wistar rat were choosed at random,then killed after3 days theywere feed. plasma and tissues were collected right after execution and were storedin -70℃ refrigerator and liquid nitrogen separately for later use . tissues ofparietal lobe ,temporal lobe ,frontallobe ,cerebellum ,mesocerebrum ,brainstem ,thalamencephalon ,hypophysis cerebrihippocampus were fetched to test ADA and XO activity by reagent boxrespectively ,then extracted the total RNA respectively by trizol for RT-PCR.ADA is the key enzyme in purine nucleotide catabolism,which can makeadenosine put off amine . ADA activity test indicated that enzyme activity inparietal lobe ,temporal lobe ,brainstem was high,the result of RT-PCR indicatedthat the relative contents of ADAmRNA in parietal lobe ,temporallobe ,brainstem was high.The result of activity test was accorded with that ofRT-PCR.XO is another key enzyme in purine nucleotide catabolism, which canchange xanthine to uric acid . XO activity test indicated that enzyme activity inparietal lobe, temporal lobe ,brainstem was high,the result of RT-PCR indicatedthat the relative contents of XO mRNA in parietal lobe , temporallobe ,brainstem was high.The result of activity test was accorded with that ofRT-PCR.The result of RT-PCR on μ-R indicated that the relative contents of μ-R mRNAin mesocerebrum , brainstem ,hypophysis cerebri,hippocampus was high and therelative contents of μ-R mRNA in frontal lobe is low .
