Objects To study the influence of virus strain multiplyof infection (MOI) and fetal bovine serum (FBS) concentration inmaintain medium on production of live attenuated mumps vaccine.Methods: We tested the virus titer in mumps vaccine which wasprepared with different MOI, and observed the influence of differentinfection dosage on production of live attenuated mumps vaccine. Weused maintain medium with different FBS concentration, 1% and2.5% respectively, to prepare mumps virus. The effect of differentFBS on production of live attenuated mumps vaccine was alsoobserved. Results: The virus titers in live attenuated mumps vaccinewere 5.76, 6.13. 5.79Log CCID50/ml with MOI 1:50,1:500,1:5000 respectively. The concentrations of live attenuated mumpsvaccine were 12.33% and 30.99%, with 1% and 2.5% FBS in maintainmedium. Conclusion: The virus titer in vaccine is highest when MOIis 1:500. Increase of FBS concentration in maintain medium willimprove the virus titer in live attenuated mumps vaccine.
