Objective To examine the effect of interaction between monocytes and vascular endothelial cells on MT1-MMP mRNA expression in high glucose conditions.Methods The monocytes were isolatied from healthy adults by centrifugation in density gradient. Monocytes and endothelial cells(EVC-304) were cultured alone or cocultured or separate cocultured to form different culture conditions,then cells were exposed to 5.5mM glucose(NG), 30.0Mm(HG) for 24h.After that, monocytes were removed from endothelial cells.The MT1-MMP mRNA expression is measured by RT-PCR.Monocytes and ECV-304 were cocultured in NG or HG medium for 24h.Then monocytes adhere to EVC-304 were determined.Results (l)There were no expression of MT1-MMP in single cultured monocytes and in separate coculturesd monocytes exposed to NGThe expression of MT1-MMP in monocytes cocultured with endothelial cells exprosed to HG and NG and in monocytes separate cocultured exposed to HG were increased significantly. The expression of MT1-MMP mRNA in monocytes of coculture system exposed to HG was most obvious (P<0.05) .(2) The expression of MT1-MMP in single cultured endothelial cells under HG reduced compared to control groups(P<0.05). Compared to the single cultured endothelial cells,the expression of MT1-MMP were increased significantly in cultured endothelial cells of coculture and separated coculture under HG(P<0.05).The expression of MT1-MMP in endothelial cells of direct coculture under HG were significantly higher than those of separated coculture under HG. The MT1-MMP expression in endothelial cells of three groups exposed to NG were not significantly different. (3) Treatment with HG for 24h significantly increased monocyte adhesion to endothelial cells (P<0.05) .Conclusion (1) HG had no effect on the expression MT1-MMP in single cultured moncytes.HG downregulated the expression of MT1-MMP in single cultured endothelial cells.(2) HG treatment significantly increased the expression of MT1-MMP in monocytes and endothelial cells that separated cocultured and cocultured. (3) HG treatment can increase monocytes adhesion to endothelial cells.
