Objective to study the protective effect of Okam J201 on bleomycin induced pulmonary fibrosis in rats and its mechanisms.Methods 64 Wister rats were randomly divided into four groups: control, model, low dose (50mg/kg , treatment groupl) and high dose (100mg/kg , treatment group2) , with 16 rats in each group. Pulmonary fibrosis was induced by intratracheal instillation of bleomycin , while the control group was given normal saline in the same condition. From then on , the rats received daily Okam J201 50mg/kg BW in the low dose group, 100mg/kg BW in the high dose group by intragastric administration , while the other rats were given normal saline daily. On the 7th day and the 28th day 8 rats in each group were sacrificed and the lungs were isolated after intratracheal instillation. Histology changes of the lungs were evaluated by Hematoxylin and eosin (HE) stain and Masson stain , and the immunohistochemical techniques were used to investigate the changes of Fas , FasL MMP-9, TIMP-2.Results Alveolitis of two treatment groups was improved significantly as compared to that of the model group on the 7th day and 28th (P<0. 01) . On the 7th day the difference of pulmonary fibrosis is significantly between model group and control group (P<0. 01) , but there was no significant difference between two treatment groups and model group or control group (P>0. 05) .On the 28th day pulmonary fibrosis of two treatment groups was improved significantly as compared to that of the model group (P<0. 01). The expression of MMP-9, TIMP-2 was significantly lower in two treatment groups than those in model group on the 7th day (P<0. 01) . On the 28th day, the expression of MMP-9 has no significant difference among four groups (P>0. 05 ), but the expression of Fas, FasL, TIMP-2 were significantly lower in two treatment groups than those in model group (P<0. 01) . Both alveolitis and pulmonary fibrisis were all no difference between treatment groupl and treatment group2 (P>0. 05) , and the expression of Fas FasL, MMP-9 TIMP-2 was also no difference (P >0.05) .Conclusions Okam J201 can alleviate alveolitis and pulmonary fibrosis, and inhibite the expression of Fas, FasL, MMP-9, TIMP-2 of pulmonary fibrosis induced by bleomycin in rats, suggesting its possible anti-pulmonary fibrosis...
