Objective:To prove long hyperglycemia resulting in rat CD59 glycation by developing a rat model, to explore molecular pathological mechanism between human hyperglycemia and vascular complications, to give an important theoretical foundation for preventing and curing diabetic vascular complications.Methods:Wistar rats(n=30) were randomly divided into three groups:the model,the group fed on a diet enriched in fat and sugar ,the normal group.After fed on a diet enriched in fat and sugar to induce insulin resistance, the rats were injected intrapertoneally with a low-dose streptozocin(STZ) to induce hyperglycemia. When all rats blood glucose level reach beyonged 16.5mmol/l and urinoglucose reached +++------++++, the successful model was made.Take rats liver,kidney and pancreas.Embed them and slice tiusse. Detect CD59 DNA in situ hybridization and detect CD59 antigen by using immunohistochemical method.Results:There were 78 section preparations of liver,kidney and pancreas, divided into (1)15 from the normal group, (2) 27 from the group fed on a diet enricheds in fat and sugar, (3)36 from the model group. (2) and (3) appeared positive (cell nucleus was blue or purplue.). (1) appeared negative (cell nucleus was not coloration.). The result of detecting CD59 antigen by using immunohistochemical method was that (2) and (3) appeared positive, but they were less than (1). All section preparations of pancreas appeared negative.Conclusion: After fed on a diet enriched in fat and sugar to induce insulin resistance, the rats were injected intrapertoneally with a low-dose streptozocin(STZ) to induce hyperglycemia. The successful model has been made.It has been proved thatthere were position mutant rat CD59 after glycation by detecting (2)and (3) in situ hybridization. It has been proved that CD59 from (2) and (3) was less than that from (1) by using immunohistochemical method. This said that CD59 glycation had no function of the normal CD59. This may help people develop theoretical foundation of the diabetic vascular complications.
