Because they are closely related to clinical disposal and therapy of diseases, and can increase the accuracy of the disease diagnosis. Thymosin beta is a newly discovered special substance which can stimulate cell movement. Thymosin beta is highly expressed in many tumor tissues and the study have already discovered that the higher expression was related to latent metestasis positively. Lots of domestic and international scholars have studied on the expression of tumor indicators in carcinoma of the breast , such as VEGF,mmp2 etc. But have little study on the expression and relationship between thymosin beta and carcinoma of the breast . This experiment have studied on the expression of thymosin beta in carcinoma of the breast and the tissue next to the carcinoma by the means of immunohistochemistry.Materials and methods1. Materials72 cases of carcinoma of the breast tissues , all from the patients underwent surgical resection in the Tumor Hospital of LiaoNing province between 2000 to 2002. At the same time, take 30 of the tissue next to the carcinomar as contrast. The tissue is fixed by 4% paraformaldehyde, embedded by paraffin.2. Clinical and pathological filesAccording to WHO international tumor organization classification is classi-fied;infiltrating carcinoma was 45 cases, ductal carcinoma was 27 cases, 3leases lymphatic node metestatic carcinoma, 21 cases haematic metestatic car-cinomar. The age is between 30 to 76 years old, the average is 50.74 years old. All these cases are not have chemotherapy and radiate therapy before operation.3. Main reagents and sourceThe primary antibody anti -human thymosin beta 4^10^15 were provided by E. Lvaniov in Greece Immunopeptide Chemistry Lab "NCSR DEMOKRI-TOS".4. MethodsImmunohistochemistry: the protein expression of thymosin beta4 A thymosin betalO and thymosin betal5 in 72 cases of carcinoma of the breast tissues and 30 cases of the tissue next to the carcinoma were examined by the means of S - P. and PBS was used as negative controls. Results judging: red in cells' is considered as positive. Because the positive rate of thymosin beta4 >,thymosin betalO and thymosin betal5 were all higher than 85 percent, cases defined as positive were regarded by both staining intensity and the percentage of immunoreactive tumor cells.5. Statistical analysisUsing Chi — square test: 0. 01 < p <0. 05 , difference was significant;p^ 0.01, difference was remarkably significant.ResultsExpression of thymosin beta4 ^thymosin betalO and thymosin betal5 in carcinoma of the breast and the tissue next to the carcinoma.In 72 cases carcinoma of the breast tissues , the expression of thymosin be-ta4 Nthymosin betalO and thymosin beta 15 mainly located in tumor cells'cyto-plasmic, in the state of diffusion. In lymphocytes and fibroblasts could find the expression of thymosin beta4 and thymosin betalO. No expression of thymosin betal5 in stroma. In 30 cases of the tissues next to the carcinoma , thymosin be-ta4 was low expressed in alveolus epithelium, thymosin betalO was slightly or on expressed in alveolus epithelium. The expression difference was remarkably be-tween carcinoma of the breast tissues and the tissue next to the carcinoma (p < 0.01).The expression rate of thymosin beta4 . thymosin betalO and thymosin be-tal5 was 86.11% (60/72), 81. 49% (59/72) , 72. 22(52/72) respectively. The expression difference had no ststistic significent in carcinoma of the breast tissues. The expression of thymosin beta4.thymosinl0.thymosinl5, in lymoph-node metastasis was 96. 67% (29/30) .95. 33% (29/31) .83. 87% (26/31) while the non - metastasis was 78. 05% .73. 17 (30/41) .63. 41 (26/41). The expression of these three proteins have remarkably difference between the ly-mophnode metastasis and the non - metastasis (P <0. 05). The expression of the proteins was relative higher in carcinoma of breast which cerbB - 2 was positive than which cerbB - 2 was negative, but there was no remarkably difference . None of these three proteins was correlated with age.ConclusionsThe difference between the expression of thymosin beta4 .thymosin betalO and thymosin betal5 protein with carcinoma of the breast lymphatic node had statistic significant. But none of these three proteins was correlated with age, sex and tumor type. |
