PART I : The protective effect of Ghrelin in severe acute pancreatitis in rat Abstract Objective: The aim of present study was to investigate the protective effect of ghrelin administration on the development of severe acute pancreatitis in rat and its mechanism. Methods: Forty-eight Wister rats were randomly divided into 3 groups i e, sham operation group, SAP model group, Ghrelin treatment group. A modified traditional intrapancreatobiliary duct injection method was adopted using Sodium taurocholate and microsyringe to make SAP models. 6h, 12h, 24h after cessation of Sodium taurocholate injections the rats were killed: detect the level of serum amylase, TNF-a and IL-6, take pancreas tissue to evaluate the existence of pathological changes of pancreas and water content of pancreas.take small intestine to evaluate the transit index. Results: Sodium taurocholate led to the development of severe acute pancreatitis. After treatment with ghrelin 6h, attenuated the development of pancreatitis. Administration of ghrelin reduced inflammatory infiltration of pancreatic tissue .Also, serum amylase, TNF-a and IL-6, water content of pancreas were reduced. But that was without effect transit index. After treatment with ghrelin 12h , attenuated the development of pancreatitis . Administration of ghrelin reduced inflammatory infiltration of pancreatic tissue .Also, serum amylase, TNF-a and IL-6 were reduced. But that was without effect water content of pancreas and transit index . After treatment with ghrelin 24h , did not attenuate the development of pancreatitis . Administration of ghrelin reduced TNF-a and IL-6 . But that was without effect water content of pancreas, inflammatory infiltration of pancreatic tissue, serum amylase, transit index .Conclusions: (1) Administration of ghrelin attenuates pancreatic damage inSodium taurocholate -induced pancreatitis;(2) Protective effect of ghrelin administration seems to be related the inhibition in inflammatory process .
