Objective: To detect the expression of EPO and EPOR in the bladder transitional cell carcinoma (TCC) tissue , normal bladder tissue and T24 cell line. Study the biologic effect of rHuEPO to bladder cancer T24 cell line and xenografts of T24 cell in nude mice. Methods: Immunohistochemistry and RT-PCR analyses were used to detect the expression of EPO and EPOR in bladder TCC tissue, the adjacent benign tissue, normal bladder tissue and T24 cell line. MTT and FCM assay were used to detect rHuEPO's effect to T24 cell's proliferation and inhibition ratio due to Hydroxycamptothecine (HPT). Form bladder cancer T24 cell xenografts, detect rHuEPO's effect to xenografts's microvessel density (MVD) and expression of the antiapoptosis protein Bcl-X_L by immunohistochemistry. Results: The expression of EPO and EPOR in bladder TCC tissue and T24 cell were higher than normal bladder tissue( p<0.01),and positively correlated with the malignant degree( r,=0.329, p<0.05). Hypoxia could induce higher expressin of EPO and EPOR mRNA in T24 cell line.rHuEPO could enhance T24 cell's proliferation limitedly,and inhibit apoptosis of T24 cell induced by HPT .It could increase the xenografts's grow up , stimulate angiogenesis and expressin of Bcl-XL-Conclusions: EPO could enhance bladder cancer cell's proliferation and inhibit their apoptosis.Use rHuEPO may promote the cancer's progression and bring negative effect in clinical practice.Which requires futher clinical reseach to prove it.
