| The mixture of Coptidis Rhizoma and Evodis Rutaecarpa is used frequently in Chinese Traditional Medical. Many preparations are made as different proportion of the Mixture. They are used directly as medical materials, which are often concluded with effective, useless, even harmful components. The quality control is absent. Both disadvantages are unreasonable for medicine. It's necessary to optimize the extraction process and establish accurate method of quality control for the Mixture. It has been proved that the alkaloids are the effective components in the Mixture. In Chinese pharmacopoeia, Berberine, Palmatine, Jatrorrhizine are selected as quality control components for Coptidis Rhizoma, Evodiamine and Rutaecarpine for Evodis Rutaecarpa. There have been many researches of extraction and purification for Coptidis Rhizoma. So the thesis concentrates on the extraction and purity process for Evodis Rutaecarpa and the quality control for the Mixture of Coptidis Rhizoma and Evodis Rutaecarpa.In the thesis, circumfluence extraction and Super Critical Extraction (SFE) were used for the extraction of the effective components such as Evodiamine and Rutaemine from Evodis Rutaecarpa. The operating parameters such as temperature, time, preassure and so on were optimized. Compare the circumfluence extraction and SFE. It was found that the alkaloids rate is higher and the extraction is vast for the former and contractive for the later. The selection is provided for the extraction of Evodis Rutaecarpa.In the thesis, both the moving characteristics of two distinct alkaloids (quaternary and indole) and electroosmotic flow (EOF) were researched in different conditions by capillary electrophoresis. On the Basis, a method of simultaneous determination of the 5 alkaloids including Berberine, Palmatine, Jatrorrhizine, Evodiamine and Rutaecarpine was developed. The separation time is less than 20 min. The method is accurate and simple. It can be used for quality control of the mixtures and the related preparations.We tried to find the purification method of the effective fractions including Evodiamine and Rutaecarpin and so on. Selecting several macroporous resins to study the adsorption and desorption function of Evodiamine and Rutaecarpin. It's found that the differences are small. The adsorptions of D101 and HPD600 are more relatively. What's more, the inference of pH value can be almost ignored. Evodiamine can be desorped by 90% alcohol, but Rutaecarpin almost can't.The addition of acetone do well for the desorption. The mixture of 90% alcohol and acetone (8:2 V/V) as the desorption solvents, the purity of Evodiamine and Rutaecarpin is 15%. The research is the base for the progressive purification of the two alkaloids. |
