Abstract/Summary:
Objective: Lung cancer is one of the most common malignant cancers known to mainland in China because of the high incidence and the high mortality.Up to now a pulmonary resection is clearly a major intervention for lung cancer.The overall 5-year survial rate is 33% in China.The uncontrolled proliferation of carcinoma cells involves an activation of oncogenetumor-suppressor gene abnormalities, and the arising,development of lung cancer has strong relationship with smoking.The high incidence and low survival arouse the biomolecular studies of lung cancer,so as to improve the prognosis early diagnosis and early treatment.It is shown that the formation and development of lung cancer is a complex progress which includes the defection and rearrangement of chromosome segments in celluar level,activation of oncogenes,deactivation of tumor depressing genes and depression of wither genes in molecular leveal. Recent studies show that multidrug resistance gene play the most significant obstacle role in the lung cancer treatment,especially in the non-small celllung cancer.Even the small cell lung cancer,which is more sensitive to chemotherapy others can appear drug resistance after several courses of chemotherapy,that leads to chemotherapy failure. By way of detecting the MDR gene expression level in non-small cell lung cancer tissue , this study aims to analyse theexpression and significance of MDR gene in non-small cell lung cancer tissue and normal tissue,the relationship between smoking and lung cancer,to provide the theoretical basis of smoking control,prevention,early diagnosis and genetic treatment of lung cancer.Methods:36 cases of lung cancer patients were selected randomly,samples of lung cancer and normal lung tissues were collected during operation,then stored freshly in -80℃ freezer and fixed in 10% Formalin solution separatedly.For each patient,clinical history collected in detail,which includes smoking history before operation and the pathological characteristics after operation,then comparison were made according to smoking history and pathological characteristics,using immunohistochemistry S-P method to detect the MDR gene expression in lung cancer tissue and normal lung tissue,analyse the relationship between MDR gene expression and clinical pathological characters, clinical pathologicalstage,cell differentiation rate,,lymph node metastasis and smoking.According to the data category,choose Chi-square to test examinate the difference of two groups,apply Spearman rank correlation analyze the related trend between different group,Kaplan-Meier estimate of survival,and combine the Log-rank test to examine the difference of two groups.Statistics analysis was proceeded through the application SAS6.2 statistics software package and p-values less than 0.05 were regarded as statisticl significance.Results:(l)The expression level of MDR1 gene in lung cancer tissues is obviously higher than in the normal lung tissues.(p<0.05)(2)The expression level of MDR1 gene in smoking lung cancer tissues is obviously higher than in non-smoking lung cancer tissue (p<0.05)(3)The lymph node metastasis have no significant influence on MDR1 gene expression in lung cancer tissues.(p>0.05)(4)The different pathological stage have no significant influence on MDR1 gene expression in lung cancer tissues.(p>0.05)(5)The cancer cell differenation have no significant influence in MDR1 geneexpression in lung cancer tissues.(p>0.05)ConclusionrThe abnormal expression of MDR gene in non-small cell lung cancer is related to cancer cell the formation development and the smoking history,has no relationship with the pathological stage,cell differenation and lymph node metastasis.
