Objective: To observe the expression of PTEN in hypertrophied hearts of spontaneously hypertensive rats(SHR) and the effect of valsartan on it, to primarily investigate the possible role of PTEN in the progression of cardiac hypertrophy in spontaneously hypertensive rats.Methods: Twenty 12-week-old male SHR were randomizedly divided into 2 groups with : SHR positive control group and valsartan treating group (30mg ·kg-1 ·d-1 ).Ten homogenous WKY rats served as normal control group. Blood pressure of rats was monitored periodically. After 8 weeks, the ratio of left ventricular weight to body weight (LVW/BW) was determined. Expression of PTEN in cardiac myocytes was examined by immunohistochemistry, expression of PTENmRNA in heart was studied by RT-PCR.Results: Blood pressure, LVW/BW were supressed markedly in valsartan group compared with SHR control group. Compared with that of SHR control group, cardiac level of PTENmRNA expression in valsartan group were significantly increased, but remained lower than that in WKY group.Conclusion: PTEN plays a role in the progression of cardiac hypertrophy in spontaneously hypertensive rats, increasion of PTEN expression could inhibit the cardiac hypertrophy, the effects may be related with angiotensin Ⅱ.
