Objective:To investigate the association among Resistin,FFA and Insulin Resistance,Fatty liver and Type 2 Diabetes.Methods:51 normal 6-week old male SD rats were divided into five groups.They were control group,fatty liver,metformin-treated,rosiglitazone-treated and diabetes group.The groups were given normal chow,high-fat diet,administration and intraperitoneal injection STZ respectively.Insulin sensitivity was determined by using insulin tolerance test.After feeding for 14 weeks,plasma and fatty tissue culture fluid resistin,serum free fatty acid(FFA),insulin,glucose,triglyceride(TG)and cholesterol total(TC) were measured in all the rats.The results were analyzed by independent-samples t test,one-way ANOVA and multiple correlation. Results :1.We succeed in bring about fatty liver,type 2 diabetes rats model through high-fat diet.2.We calculate the descent width of blood glucose with Nor-sugar rate,as insulin tolerance test showed,the nor-sugar rate of fatty liver group was only 66% of control group's and prove their severe insulin resistance.Rosiglitazone significantly increased nor-sugar rate by about 50% and Metformin 15%,diabetes group increased about 26% when compared with the fatty liver group.3.The plasma resistin of the rest groups showed elevated tendency as compared with the control group,but ANOVA showed no significant difference among them.4.The FFA,TG,TC of fatty liver group have no apparent chang compared with that of control group.Rosiglitazone and diabetes caused an reduction of FFA(P<0.05).Rosiglitazone decreased TG by 23%(P<0.001) and TC level by 7.0%(P<0.01),Metformin decreased TG by 17.2%(P<0.01) and TC level by 9.3%(P<0.01)when compared with the fatty livergroup.5.The empty stomach insulin level of diabetes group is lower 33%(P<0.001),fasting blood glucose is higher 100%(P<0.001) and insulin resistance index is higher(P<0.01) as compared with the rest groups.The three index among the rest groups showed no statistics difference(P>0.05).6.The plasma resistin of all groups have no linear correlativity to nor-sugar rate,FFA,insulin resistance index,insulin,blood glucose,TG,TC(P>0.05).The TG and blood glucose of diabetes group is correlated,and coefficient correlation is 0.812(P<0.001). Conclusion: 1.14-week high-fat diet induces severe insulin resistance in normal SD rats,Rosiglitazone and Metformin treatment ameliorate IR in a great extent and are accompanied by a decrease in TG and TC level;2.H0MA insulin resistance index is unsuitable to evaluate IR degree of rat.3.Resistin have no correlativity to fatness,IR,Fatty liver and Type 2 diabetes of rat,and isn't influenced by the treatment of Rosiglitazone or Metformin.
