OBJECTIVE To investigate the relation between cytokines derived from keratinocyte exposure to UV and the disbalance of T helper cell subgroup. To offer theoretical and experimental evidences for the further studies the pathogenesis of photodermatitis.METHODS Using different dose of UVA and UVB to irradiate HaCaT cell line respectively.At the time of 12h after irradiation ,observed the expression of IL-10R, IL-4R, IL-12R and IFN- Y R in HaCaT cells by immnohistochemistry(S-P method) and calculated the percentage of positive cells. At the same time, quantified the protein levels of IL-10, IL-4, IL-12 and IFN-γ in supernant of irradiated cells with ELISA method.RESULTS 1. Without irradiation, there were low expression of IL-10R,IL-4R, IL-12R and IFN-γR in HaCaT cells and there were no or low levels of IL-10, IL-4, IL-12 and IFN-γ in supernants. 2. There were no or little expression of IL-12R, IFN-γR in the HaCaT cells and the same case was seen in supernants 12h after UVA and UVB irradiation. 3. With UVB irradiation, upregulation of IL-10R and IL-10 was detected 12h after irradiation, and the similar situation was observed in lower dose irradiatin of UVA. In contrast, the higher dose of UVA down-regulated the expression of IL-10R and IL-10. 4.The expression of IL-4R in the HaCaT cell was up-regulated in the group of higher dose of UVA and UVB;There was no change with IL-4 secretion in supernant and no IL-4R expression after lower dose UV exposure.CONCLUSIONS 1.The effects on keratinocyte-derived cytokines after UV exposure are different according to the dose of irradiation and the kindof cytokines. 2. The keratinocyte-derived cytokines after exposure to UV can cause disbalance of Thl/Th2. 3. Our results show that:the disbalance of Thl/Th2 caused by keratinocvte after UV exposure probably play an important role in the pathogenesis of potodermatitis. Of course, the further experiment is necessary.
