Objective To study the effect of portal vein occlusion on intestinal mucosa barrier in rats and the protection of ulinastatin to the injury, To present the experimental data for the clinical surgery. Materials and Methods 70 Sprague-Dawley rats were randomly divided into controlled group, operation group and operation and medication group .The portal vein were occlused 30 min, 40 min, 60 min respectively in the operation groups and operation and medication groups. All the rats were killed 5 hours after operation.And 2ml blood from portal vein , lymph nodes around appendix, 1cm small intestine wall were taken for endotoxin levels, bacterial translocation and pathology examinations. The mocusa barrier and microscopic structure of intestine were observed. Results Compared between the control group and the operation group ,endotoxin levels, bacterial translocation rates rise greatly and gut structure change obviously in the later. The above change become more obvious with the increase of portal vein occlusion time. Compared between the the operation group and operation and medication group, the above changes is also obvious in the later. With the time increase,the difference become smaller. Conclusion The occlusion of portal vein can lead the decrease of intestine mocusa barrier and the increase of its permeability. There is a positive correlation between the above change and the time of portal vein occlusion. Ulinastatin has a good protective effect on the above damages.
