Objective: To investigate the effects of panax notoginseng saponins(PNS) Rg1 on the expression of bFGF protein in rats following cerebral ischemia-reperfusion.Methods: 60 adult male SD rats were divided randomly into model group, positive control group and PNS Rg1 group. Based on different doses, PNS Rgl group were divided into Rg1 low-dosage group, Rg1 med-dosage group and Rgl high-dosage group, and based on different reperfusion time, each group was divided into 1d-reperfusion group, 3d-reperfusion group and 7d-reperfusion group. Each group had 4 SD rats. The ischemia-reperfusion model of middle cerebral artery were set up. According to the difference of the model group, the positive control group and PNS Rg1 group, the rats were injected with 0.9% Nacl 5ml/mg, Nimodipine lmg/kg, Rg1 50mg/kg, 100mg/kg, 200mg/kg in abdominal cavity, one time a day. The rats were sacrificed at the first day, the third day, and the seventh days. Then the cerebral tissue was made into frozen section in 14μm thickness. The sections were identified under the same condition using specific bFGF(1:100) antibody by immunohisto-chemistry ABC method. The positive bFGF neurons in cortex were counted. All were analyzed by one-way ANOVA and q test.Result: 1. The expression of bFGF was seen in the different groups. 2. On the 1st day, the expression of bFGF in the different groups had no significant difference (P>0.05). On the 3rd day and 7th day, the expression of bFGF in the model ,the positive and Rgl group had significant difference (P<0.05) . The expression of bFGF of Rgl group at the different time had no significant difference (P>0.05) .Conclusion: 1. The expression of bFGF in normal and ischemic cerebral cortex has some effect to maintain and recovery to neurons. 2. For cerebral ischemia-reperfusion, Nimodipine and PNS Rgl can protect and treat brain, and can sustain at high level expression of bFGF, and can stave the descending speed of bFGF 3. PNS Rgl have the function of treat and protect when it was used in local ischemia, but it has no dose-effect relationship from experiment. However, it shows evident time-effect relationship. The time of treatment can not be too short, seven days is the best.
