Objective: to investigate the protective effects and mechanism of Fleeceflower Root on the cultured anoxic damnification cardiac muscle cells in vitro by the methods of histochemistic and biochemical measurement.Methods: it was cultured the cardiac muscle cells of wistar suckling rats for 5 days, obtained the well-grown cultured flasks and divided into 4 groups: normal group, model control group, medicine control group(Panax Saponin) and experimental group(Fleeceflowev Root). Cells of normal group was cultured in mormal culture medium. Cells of model control group was cultured in N2 saturated anoxic medium. Medicine control group was cultured in medium with N2 saturated and 0.05% Panax Saponin. Cells of experimental group was cultured in medium with N2 saturated and 0.5% extraction of Fleeceflower Root. After cultured continously in the CO2 thermostat at 37℃ for 12h, Hepatin was show with PAS method. ACP and SDH was show by histochemistic methods. The activitiy of SOD and content of MDA were detected by biochemical methods.Results: in the normal group, PAS reaction of cardiac muscle cells was strong; theamout of the hepatin granule was dense; the activity of SDH was strong; the amout of the blue-purple granule in the cytoplasm was dense; the activity of ACP was slight; the amout of the brown granule in the cytoplasm was less and the coloration was slight. In the model control group, PAS reaction of cardiac muscle cells was slight; the amout of the hepatin granule was decreased significantly and the coloration was slight; the activity of SDH was decreased; the amountof the blue-purple granule in the cytoplasm was decreased significantly and the coloration was slight; the activity of ACP was increased; the amount of the brown granule in the cytoplasm was increased and the coloration was dense. Compared with model control group, experimental group and medicine control group PASreaction was immmd significantly; the amount of hepatin granule was increased and the coloration was strong; the activity of SDH was increased significantly; the amount of blue-purple granule in the cytoplasm was increased significantly and the col?ra|ion was strong; the activity of ACP was decreased; the amount of brown granule in the cytoplasm was decreased and the coloration is slight Compared of normal groups the activity of SOD was decreased in the cells of the model control group and the content of MDA was increased significantly(P<0.01). Compared of model control group, the activity of SOD was increased and the content of MDA was decreased sigftif|cantly(P<0,01).Conclusions: the Jesuits prove that Fleeceflower Root possessed a very well protective and anti-oxidation effects on the anoxic cardiac cell.
