Objective: 1.The purpose of the first study was to assess  bone marrow stromal cells(BMSCs)and/or platelet-rich plasma(PRP) auto-transplanted to promote periodontal regeneration in canine experimental classⅡfurcation defects. 2.To compare the difference in promoting periodontal regeneration of initial cell seeding density in canine experimental class Ⅱfurcation defects . Methods: 1.A total of 30 experimental classⅡfurcation defects were randomly divided into four groups and were surgically repaired by GTR,GTR/PRP/Bio-Oss,GTR/BMSCs and GTR/BMSCs/PRP/Bio-Oss.The animals were sacrificed at 12 weeks postsurgery for histologic and histometric analysis. 2.GTR combination with the expanded BMSCs which were seeded in collagen membrance at initial concentrations of 5×105cells/ml,5×106 cells /ml or 5×107 cells /ml ,and auto-transplanted into experimental class Ⅱfurcation defects,underneath a Bio-Gide membrane. Bio-Gide membrane alone was implanted into the defects as a control.Periodontal regeneration was evaluated by histologic and histometric analyses 12 weeks after transplantation. Results: 1.Significantly greater periodontal regeneration was noted in the groups of PRP/BioOss/ BMSCs /GTR compared with the PRP/BioOss/GTR, BMSCs /GTR, GTR treatment alone when newly formed periodontal tissues including alveolar bone, periodontal ligament and cementum were assessed. Interestingly, no significant difference was found between GTR alone or plus BMSCs  or GTR combine with of PRP/Bio-Oss. The amount of new alveolar bone formation, new cementum formation, new periodontal ligament tissue formation in the PRP/BioOss/ BMSCs /GTR group was 1.75±0.840mm,2.271±0.704mm,2.606±0.410mm,  respectively. The GTR group: 0.737±0.487mm,1.149±0.675mm,1.514±0.905mm; the PRP/Bio-Oss/GTR group: 0.494±0.564mm,1.414±0.638mm,1.557±0.772mm; the GTR/ BMSCs group: 0.82±0.576mm,1.657±0.619mm,1.871±0.670mm. 2. The percentage of new cementum length in the 5×105cells/ml,5×106 cells /ml and 5×107 cells /ml groups were significantly higher than in the control group (P<0.01), and there was no significantly difference in the three cell groups. The percentage of new alveolar area in the 5×106 cells /ml and 5×107 cells /ml groups were significantly higher than in the control and 5×105cells/ml group (P<0.01), and there was no significantly difference between the first two groups,and neither was the later.  Conclusions: 1.The data from the first study suggest that the application of GTR combination with PRP and BMSCs,but not alone can support the periodontal tissue regernation in canine. 2.The second study suggest that the effect of auto-transplanted BMSCs promoting periodontal healing has cell density–dependent under certain conditions,and the higer concentrations(5×106 cells /ml and 5×107 cells /ml) can  effectivly promote periodontal regeneration.
