Objective:To study effect of kaixinsan on learning and impairment induced byamploid β- peptide25-35 (Aβ25-35) rats, as well as the change of expression of app, bax ,bcl—2, and Trb3 gene.Methods:(1)The experimental model of animal:Female and male Wistar rats with normal cognition are randomly divided into five groups, Aβ group, control group, low dose of kaixinsan group, high dose of kaixinsan group, and naofukang group. Each group is composed of 12rats. Aβ25-35 was injected into the rats' right Meynert nucleus in A β group, low dose of kaixinsan group, high dose of kaixinsan group and naofukang group. 0. 9%NS was injected into the right Meynert nucleua in control group. Next day, kaixinsan were given respectively by gastric canal in mouth cavity in low dose of kaixinsan group and high dose of kaixinsangroup, at the same time, naofukang was given to naofukang group by the same approach, besides, 0. 9%NS group and Aβ group were fed for 28 days. 28 days later, to detect the praxiology of these rats by Morris water maze.(2)The experiment to treat AD induced by Aβ25-35 in rat. At the end of the experiment, the rat were decapitated, then brain was separated for examination of Ache activity and theexpression of APP protein, bax, and bcl-2 by immunohistochemistry method. Last, detect the expression of bax, bcl-2 and Trb3 gene by PCR.Results:1-The effect of kaixinsan on cognition in AD rat: In Morris water maze test, A {3 group compared with control group, the avoiding latent period extended, P<0. 01;Compared with A {3 group, the avoiding latent period of low dose of Kaixinsan group, high dose of Kaixinsan group, Naofukang group shorten, P<0. 05. kaixinsan can improve the cognition in AD rat in Morris water maze test.2.The acetyl cholinesterase level of the rats brain is lower notably. The AchE activity of A {3 group is obviously enhanced, and it compared with control group, P<0. 01 o Compared with A {3 group, the AchE activity of low dose of Kaixinsan group, high dose of Kaixinsan group, Naofukang group decreased, P<0. 053.The expression of APP as well as the apoptosis protein bax can be declined by Kaixinsan, and the apoptosis protein bcl-2 can be enhanced. 4.The expression of the bax and Trb3 gene was restrained by Kaixinsan.
