Pueraria Lobats is a kind of leguminous plant' radix. It distributes in most districts of Chins and grows in warm and moist surroundings. It can be divided into puenaria lobata (wild) Ohwi and pueraria thomsonii Benth. Pueraria Thomoni benth contains rich starch, which is a kind of important source of food and additive. Puenaria lobata (wild) Ohwi contains rich isoflavones, polysaccharide, amino acid and microelements etc. Isoflavones have significant pharmaccutical functions and pharmaccutical value and behave to to human health.Isoflavones is an important kind of scarce raw materials in medicine and chemical industries. Radix puerairia contains rich isoflavones. Pueraria Lobata is a kind of rapidly growing plants, which is suitable for extensive implantation in Lianyungang. However, pueraria is only used to extract starch. And the most isoflavones are discarded. Flavnonoids are eagerly needed in either domestic or the international markets. The extraction and purification techniques are needed to improve at present. This paper systematical analyzed the main component of the pueraria of Lianyungang. And detailed studies t he p rocess o f e xtraction a nd p urification o f t he i soflavones from Lianyungang radix pueraria. The major results were as follows:1. Eliminated the solution with inductively coupled plasma atomic emission spectrometry (ICP-AES), which is used to determine inorganic elements. With the mixture acid to eliminate the solution directly, the determination results indicate that Pueraria Lobats of Lianyungang contains many inorganic elements such as K, Ca etc. The content of Ca is 7.94μg/mg. The contents of Fe, Mn, Zn, and Cr are 0.170μg/mg, 0.0190μg/mg, 0.0253μg/mg, and 0.500μg/g respectively. The content of poisoning element As is zero.2.Adopt the spectrophotography to analyze total sugar of Pueraria Lobats. Abstract the sample of Pueraria Lobats with the alchol and the benzene anthranol- sulfuric acid as the developer. The result shows: The contents of polysaccharides in Pueraria Lobats are very difference between different sorts and places. And the Lobats wild in south are higher than that o f in north, Lianyungang 1 obata Ohwi is 23.1%; Lianyungang wild is 10.3%.3.Extraction isoflavones from Pueraria Lobats.The single factor experiments showed that pH value, temperature, solvent and concentration of solvent influenced the macerating more. The optimal conditions were pH value of solution was 6~8. The temperature was 85℃. The concentration of alcohol was about 70%.4. The optimal macerating conditions were obtained with the orthogonal experiments that 70% alcohol (V/V) as extraction agent, 85℃, pH6-8, 1 h, 2 times, the ratio of solid/liquid 1:10. The ratio of solid substance macerating, the content of products and the macerating efficiency were 19.9%, 42.6%, and 92.3% respectively.5. By hydrolyzed of n-butyl alcohol extract with 5% HC1, the extract efficiency increased a little. After using n-butyl alcohol extracted for four times, the pueraria isoflavones was almost completely extracted. The yields and the purity were up to 50.9%and 81.0% respectively.6.The o ptimal c rystallization c onditions o f p ueraria i soflavones w ere : 10°C, 9 5% consistency of ethanoic acid solvent. The ratio of solid/liquid was 1:1.5. The yield and the purity were 36.8% and 85.5% respectively.7.A column chromatographic determination was made for the separation of isoflavones. The solid-phase was silica-gel, the eluting reagent was CHCI3-CH3OH- H2O (4:i:0 05). By HPLC detection ,the content of puerarin was up to 95.2%.8.We carried out the spectral analyses IR/H-NMR^C-NMRof our product.Our product was conformed to be puerarin by comparing the charts and data with that of the standard atlas and the document data.
